Color Picker Tool and Grayscale

How do I connect my wacom intuos tablet pen buttons to the color picker tool and how can I turn a layer from luminosity to transparency?

1)Just bind alt key to required button
2)What do you mean? Is this function available in photoshop?

This feature is available on Paint Tool Sai. It kinda helps to grayscale in a way but turns white to transparency.
